Frackers

  1. Found that RNA quality is good enough to run microarrays; RIN numbers look good, but still unsure if 'bad' numbers would result in poor microarray results.  Unclear what the future is with these results.
  2. Current extraction testing has been on 16s gene; must test functional genes and chris P getting p/p for rbcl, AOAa, and Roseobacter.  Hope to have two finished this week.  If these work, Chris has multitude of questions to move forward.  Looks like enzymatic lysis will not work for RNA; works great for DNA, but too slow for RNA, allowing RNAses in to destroy RNA before complete lysis.
  3. Elif has finished standard curves for pier deployment and is now crunching the numbers.

    1. almost completely cleaned.  Seems like o-rings are culprits with 10^4 DNA copies hanging out in the top and 10^3 in bottom on collection side.
    2. NEO
      1. same issues.  Roman has helped develop a cleaning protocol, and we now know where to look first when heavily contaminated.
  5. To run BASIC event detection (i.e., threshold crossing) Brent needs to test the new Ruby interpreter.  This will be installed on NEO this week after Roman fixes bearing and leak.

Particle Concentrators

  1.  Still waiting to hear from Kia.  Jim will ping him.
  2. We have settled on dead-end filtration, but may not be constrained to circular form factor, just similar surface area.
  3. Is size-fractionation still a requirment?  Yes.  it is one of our three operational-use scenario.

Cawthron Deployment

  1. Email sent to Chris Cornelisen.  Ideas:
    1. deployment will be May 1-31, 2012
    2. must get two cores to Cawthron by April 2 to have time for set-up.
    3. Kevan has sent list of targets we are proposing to go with.
    4. MBARI will be paying for reagents/assays, but Cawthron should buy directly from Spyglass to mitigate customs issues.
    5. post-deployment runs to be performed AT Cawthron.
    6. Doug developing a shipping option for the cores which will be 'dry-shipped' via air freight.
  2. We have only 1 ISUS and it will be going to NZ.

Other Items

  1.  Do we need permits to put ESP on pier in Newport Beach/Orange County?
  2. Passports--everyone check that your's are up-to-date.  Kevan has checked and Visa is not needed for New Zealand.
  3. We need some hard dates related to G3 development so people can plan their lives, and we can work development around the many deployments.
  4. Need to find out what depth Drifter needs to be at for Sept CANON cruise.  Currently have 1 25m cable.  If they want an oxygen sensor on CTD, they should buy (or we'll split it with them)
  5. We are having 4 E/M cables made.  Two 15m (finished) and two 25m (in que to be built)
